Slow-release 'jelly' delivers drugs better

Tuesday, January 29, 2013

Duke University biomedical engineers have developed a new delivery system that overcomes the shortcomings of a promising class of peptide drugs ? very small proteins ? for treating diseases such as diabetes and cancer.

There are more than 40 peptide drugs approved for use in humans and more than 650 are being tested in clinical studies. One example is the hormone insulin, a peptide that regulates the metabolism of carbohydrates in the body and is used as a drug to treat diabetes.

Despite their effectiveness, peptide drugs cannot achieve their full potential for a number of reasons. They are rapidly degraded in the blood stream and they are cleared rapidly from the body, which requires multiple, frequent injections. Because of this, peptide concentrations in the blood can rise precipitously just after injection and fall dramatically soon thereafter, causing unwanted side effects for patients.

One popular method to solve this problem involves loading peptide drugs into polymer microspheres that are injected under the skin and slowly degrade to release the peptide drug. Microsphere-release technology has proven useful, but has many issues related to its manufacture and ease of patient use, the researchers said.

"We wanted to know if we could create a system that does what the polymer microspheres do, but gets rid of the microspheres and is more patient-friendly," said Ashutosh Chilkoti, Theo Pilkington professor of biomedical engineering in Duke's Pratt School of Engineering.

The new approach involves making a "fusion protein" that consists of multiple copies of a peptide drug fused to a polymer which is sensitive to body heat. The fusion molecule is a liquid in a syringe but transforms into a "jelly" when injected under the skin. Enzymes in the skin then attack the injected drug depot and liberate copies of the peptide, providing a constant and controllable release of the drug over time.

Miriam Amiram, former Chilkoti graduate student and first author on the paper, dubbed the new delivery system POD, for protease-operated depot.

In the latest experiments, published on-line in the journal Proceedings of the National Academy of Sciences, the researchers fused glucagon-like peptide-1 (GLP-1), a hormone that regulates the release of insulin, with a genetically engineered heat-sensitive polymer to create the POD.

"Remarkably, a single injection of the GLP-1 POD was able to reduce blood glucose levels in mice for up to five days, which is 120 times longer than an injection of the peptide alone," Chilkoti said. "For a patient with type 2 diabetes, it would be much more desirable to inject such a drug once a week or once a month rather than once or twice a day.

"Additionally, this approach avoids the peaks and valleys of drug concentrations that these patients often experience," Chilkoti said.

Unlike peptide-loaded microspheres, PODs are also easy to manufacture, because the peptide drug and the heat-sensitive polymer are all made of amino acids. They can be built as one long stretch of amino acids by engineered bacteria.

"This new delivery system provides the first entirely genetically encoded alternative to peptide drug encapsulation for sustained delivery of peptide drugs," Chilkoti said.

Google maps out North Korea with crowd-sourced data

4 hrs.

While Google Maps feels like a solid mapping solution, it has some blank spots?? ?particularly when it comes to regions such as?North Korea. But that's all changing thanks to a group of "citizen cartographers" and a tool which allows Google to incorporate crowd-sourced map data into its product.

As?Jayanth Mysore, a senior product manager working on the Google Map Maker project,?explains in a post on the official Google Maps blog, the Map Maker tool?has been around since 2008 and allowed users to "update the maps of the areas they know, and improve their level of detail and accuracy." Crowdsourced data like this is how Google "will build the modern map," Mysore adds, reiterating that without these details, map data is currently "very limited" in some parts of the world.

Efforts to map out North Korea have been made over the last few years, but on Monday, Google was finally ready to officially update the region on Google Maps. "We know this map is not perfect," Mysore acknowledges. "We encourage people from around the world to continue helping us improve the quality of these maps for everyone with Google Map Maker." He adds that, from now on, any further "approved" updates to the North Korean maps will also appear on Google Maps.

The Wall Street Journal's Evan Ramstad points out?that the update of North Korea on Google Maps comes about three weeks after Google's executive chairman Eric Schmidt visited the area (though a Google spokesperson tells NBC News that the visit was unrelated to the timing of the Google Maps update).?During his time in the country, "Schmidt encouraged officials he met in North Korea to make the Internet available to its citizens and end its attempts to restrict information," Ramstad writes.?The Verge's Sam Byford reminds?that Internet access in North Korea remains quite restricted nonetheless, and that odds are that the "vast majority of North Korean citizens" won't be able to access Google's freshly updated maps.

The crowd-sourced cartography includes mass transit, monuments and parks, as well as North Korea's massive gulags, which are signified on Google Maps with a slightly different shading. ?As?the Atlantic notes, "Naturally, the Hwasong Gulag, like any place on Google, already has some jokesters reviewing its?accommodations."?

Mysore dances around these detail in his blog post, and instead focuses on who will be able to take advantage of the information. "While many people around the globe are fascinated with North Korea," he writes, "these maps are especially important for the citizens of South Korea who have ancestral connections or still have family living there."

Providing the Right Information for Call Centre Workers

Managing a call centre is about getting results, and the people on the front line making the calls need the right information if those results are to be achieved.

This information includes:

  • Sales training
  • Performance feedback
  • Product knowledge
  • Motivational training
  • Hands-on training.

Sales training

Cold calling requires a thick skin, determination, patience, a personable nature and plenty of confidence in your own ability. It also requires training and call centre operatives should receive induction training that covers the basics of the system they will be using, as well as comprehensive and ongoing sales training.

Feedback

Performance feedback is an invaluable learning tool. Call centres today have sophisticated contact management software at their disposal that lets them analyse a recorded phone conversation and determine weaknesses in an operative?s performance.

An example of this is phonetic search software that allows you to search for particular words and phrases in a conversation, which may indicate problems in the operative?s call style or approach. Playing the operative examples of successful calls is also the kind of information they need to improve.

Product knowledge

Call centre operatives need to know what they are talking about. They must be provided with detailed information about what they are trying to sell, in order to be able to convince a customer of its merits. This knowledge can be imparted through training sessions, product manuals and ? more often now, thanks to the sophistication of modern call centre software ? access to instant information they can call up on their screens.

Motivational training

Cold calling is all about percentages. While a majority of callers may say ?no?, it is the small percentage that says ?yes? that matters. Because operatives face rejection and even hostility on a daily basis, keeping them motivated is a very important part of managing a call centre team.

Teamwork and mutual support are the keys to maintaining morale and they need to become a part of your company culture. Encouraging operatives to focus on the goals and rewards and how they can achieve them will help to keep negativity at bay and staff retention rates high.

Hands-on training

Rather than being forced to sit through lectures, operatives should be given training ?in the field,? as the best way to learn something is always to do it yourself. Practical training could include role-playing, where particular scenarios are played out and the operatives? performances are discussed and analysed by the group.

Another practical training method is to allow them to listen in on your best operatives at work and have those operatives share their methods for success. A new operative with a role model or mentor to look up to and learn from will also do far better than one who is dropped in at the deep end and left to sink or swim.

Information is knowledge and knowledge is power. The best way to empower your call centre workforce is not to overload them with information, but to give them the right information that they can use to improve and ultimately deliver the results you need.

Ford posts strongest pre-tax profit in over a decade

12 hrs.

Ford Motor says it made the highest fourth-quarter pre-tax profit in more than a decade. It earned $1.7 billion last quarter, due largely to increases sales of trucks and SUVs and higher prices for all models.

That was 31 cents a share, beating Wall Street forecasts which had ranged from 25 to 28 cents.

For all of 2012, the automaker?s net income was $5.7 billion, down $307 million than a year ago. It excludes some special charges.

The Detroit Bureau: Ford, Daimler and Nissan form fuel cell alliance

?The Ford team delivered strong results once again, underscoring that our One Ford plan is working,? proclaimed Alan Mulally, Ford president and CEO. ?We are well positioned for another strong year in 2013, as we continue our plan to serve customers in all markets around the world with a full family of vehicles ? small, medium and large; cars, utilities and trucks ? with the very best quality, fuel efficiency, safety, smart design and value.?

Revenue last quarter was $36.5 billion, up $1.9 billion from the same period a year ago. For 2012, revenue was $134.3 billion, down $2 billion from a year ago.

Trucks, mainly sold in North America, drove profits hard enough to overcome European losses ? which were substantial, and more than the maker had forecast just a few months ago.

The Detroit Bureau: Toyota officially reclaims global sales crown

In the quarter, Ford reported European losses of 732 Million, against a loss of $190 million the previous year. For all of 2012 Ford?s European loss climbed to $1.73 billion. The automaker had forecast about $1.5 billion when it discussed third-quarter earnings three months ago.

In North America, Ford said it made $1.87 billion in the quarter ? twice as much as the $889 million on it posted a year earlier. Revenue for the quarter was $22.1 billion, up $2.5 billion.

For the year, North American profit was $8.34 billion, before taxes.

The Detroit Bureau: 2013 care sales off to good start

Ford showed a preview of the 2015 F-150 at the Detroit auto show earlier this month, hoping to tempt prospective buyers into waiting instead of buying General Motors new Chevrolet Silverado or GMC Sierra, on sale first half this year.

Ford also wanted to raise doubts in shoppers? minds about spending now for Chrysler Group?s Ram pickup, revised for 2013 and winner of the North American Truck of the Year earlier this month, an award bestowed by 49 U.S. and Canadian auto journalists.

Ford had reported a loss of $468 million in Europe last quarter and warned that losses were mounting. The automaker has announced plans to close three European plants by 2014, but said last quarter that it would lose $1.5 billion in 2012 and again this year, and doesn?t expect black ink from European operations until about 2015.

Amped Wireless High Power Wireless-N 600mW Gigabit Dual Band Access Point (AP20000G)

Amped Wireless' High Power Wireless-N 600mW Gigabit Dual Band Access Point (AP20000G) is a piece of networking hardware that can either be used to bring wireless access to an existing network, or to upgrade wireless networks with legacy 802.11x technology to the more modern 802.11n standard. The access point (AP) delivers excellent throughput, especially on the 5GHz band and provides very good coverage but lacks some more business-oriented features of some SMB access points on the market.

Specs
The AP20000G operates at the 2.4 and 5GHz bands. The device supports up to 600Mbps of theoretical throughput. Dual amplifiers on both the 2.4 and 5GHz wireless radios help the access point with wireless signal coverage. Two high gain 5dBi dual band antennas screw into the antennas ports on the casing.

The access point features five Gigabit Ethernet ports?one for wiring to a router or switch and four for connecting other networking devices to leverage Gigabit speeds. There's also a USB 2.0 port for sharing data on connected USB storage drives.

Users can operate the access point on an included stand, or flat on a desktop. The device is also wall-mountable.

Set Up and Configuration
There's very little to set up; the process is very easy. I powered the AP and connected its port labeled "network" to a switch on my network. I also connected a laptop to one of the ports on the AP labeled "Wired Devices."

The AP takes a few seconds to configure itself. There are several LEDs on the device's front panel that light up when power is on, when a device is connected to one of the Ethernet ports, and when there is an active network connection.

Once the LEDs were lit, I could see two new SSIDs, created by the AP20000G in my laptop's list of wireless networks. The AP creates an SSID for each band. The security key for each is printed on the bottom of the access point.

Of course, you will want to change the default SSID password and the AP's admin password. To configure the AP, I just opened a browser on my laptop and pointed to the URL: setup. ampedwireless.com. The interface has the same neat, intuitive-to-navigate design of Amped Wireless' High Power Wireless-N 600mW Gigabit Dual Band Router (R20000G).

The AP20000G shares some features that are built-in across Amped Wireless' product line. One such feature is the ability to control the wireless coverage area. Radio buttons allow you to set coverage at 100 percent (which is the default setting), down to 15 percent. Lowering the coverage area means lowering the range? area in which the AP broadcasts itself, which may be desired for security reasons, and lowers the overall output power.

Users also have the option to disable the wireless radios based on a schedule by day and time Access to the AP can be granted or denied based on a MAC address control list.

The AP is designed to extend wireless network connectivity in small business networks and homes. I would recommend it as an AP more for consumers and for smaller business networks not requiring a lot of security or controls. This is because, aside from RADIUS authentication support, there are fewer business-class features in the AP20000 than some other SMB APs we've tested. For instance, there's no native firewall or Active Directory integration, as there is in a true SMB AP such as the Meraki MR16.

While the AP20000G supports up to eight guest networks, you can't create a custom splash page for guests that access the AP as you can with the Meraki MR16?device.? However, each guest network on the AP20000G can be set with its own login credentials and you can limit bandwidth for each network.

Also, there's no AP isolation preventing wireless clients from communicating with one another when they are connected to the AP which may be a security concern for some.

These lack of features may be fine for smaller businesses especially ones with routers that have these features such already, such as a built-in firewall. However, those needing APs with more business controls may want to look into business-class APs from Meraki or HP.

I found the same lengthy reboot required for just about changing any settings in the AP20000G as in Amped Wireless' R20000G router. A settings' change requires a 70 second reboot of the device which can be slightly annoying when you are first configuring the AP the way you want.

Performance
Amped Wireless' main focus is on range. You get good range and very good throughput, in fact the top throughput we've tested for an AP at 5 GHz, with the AP20000G.

The speeds are more on par with a dual-band wireless router. In 5 GHz mode at 30 feet, the AP2000G clocked a very impressive 113 Mbps. In comparison, many of the SMB APs on the market have tested quite a bit slower. For example, Meraki's MR16 AP? also a dual-band 802.11n AP? only managed 49 Mbps at the same distance in 5 GHz mode.

As expected, the AP20000G was slower, albeit, still with decent throughput in 2.4 GHz-N only mode, achieving an average throughput of 74 Mbps. Below is a chart comparing the AP20000G with other APs at 5 GHz:

Speedy, But Biz Feature-Light
For home users looking to extend a wireless network in a coverage area where a less-powerful wireless extender wouldn't get the job done, the AP20000G is an ideal piece of networking hardware.

Although this is a fast access point with very good coverage, business networks that need more control over security and user access may need to look into access points specifically marketed to the SMB. On its own,? the AP20000G is solid hardware showcasing Amped Wireless' commitment to delivering network products that are fast-performing and able to cover large areas. The AP20000G is a 3.5 star earner and is a very good access point.

Israel's comatose Sharon showing brain activity

(AP) ? A team of Israeli and U.S. scientists say new tests on comatose former Prime Minister Ariel Sharon show significant brain activity.

Ben-Gurion University on Sunday said Sharon responded to external stimuli at Soroka Hospital in Beersheba.

He was shown pictures of his family and listened to recordings of his son's voice while undergoing a special brain imaging scan. The university said "significant brain activity was observed ... indicating appropriate processing of these stimulations."

However, Sharon remains in a deep coma.

Sharon, 84, led Israel from 2001 until suffering a stroke in 2006. Since then, he has been in a vegetative state, connected to a respirator.

Sharon was a storied military officer who fought in three wars before entering politics. He unilaterally withdrew Israeli troops and settlers from Gaza in 2005.

Space Weather and the Aurora | Royal Meteorological Society

With January coming to an end, it will only be a few weeks until the vernal equinox. In 2013 it falls on 20 March.

Spring is the time of year when there is an increase in the number of aurora, both Borealis in the northern, and Australis in the southern hemisphere.

The Aurora, a kind of "space weather" are caused by energy coming up against the Earth's magnetic field. The energy comes from the sun, which acts as an enormous power plant, with temperatures reaching in excess of 14 million C in places. On the outer layers of the sun, the energy moves in huge eddies called convection cells. These currents create electrical fields in the sun. On the surface, plasma is formed, which can then travel outwards and break away from the sun in huge pieces, forming what are known as solar storms. The image below shows the Earth to scale, against solar activity. There are different kinds of activity, including solar storms, coronal mass ejections (CMEs) and sun spots.

solar flare

A solar storm can reach speeds of over 8 million kilometers an hour. After 6 hours it can pass the planet Mercury and after 12 hours, Venus. After 18 hours the solar storm may reach Earth. When the storm reaches Earth, it comes up against our magnetic shield. This can interact with the solar energy and create a funnel, which sends the energy around the Earth and towards the poles, where it can sometimes be seen as beautiful green lights.

The sun is currently reaching the peak of its 11 year activity cycle, but it is thought that 2013 will be a year of relatively low activity.

Japan To Start Broadcasting 4K TV in 2014

The Japanese government will start 4K TV broadcast in July 2014, a move that could to help sales of ultra high-definition televisions, the Asahi newspaper reported on Sunday.

The service will begin from communications satellites, followed by satellite broadcasting and ground digital broadcasting, the report said.

Broadcasts will start using communications satellites (CS,) followed by satellite broadcasting and ground digital broadcasting, the report said.

The 4K TVs or UHDTVs boast four times the resolution of current high-definition TVs, are now on sale by Japanese makers including Sony, Panasonic and Sharp as well as by South Korea's LG Electronics and Samsung Electronics.

According to the ITU's specifications for UDTV, the first level of UDTV picture levels has the equivalent of about 8 megapixels (3,840 x 2,160 image system), and the next level comes with the equivalent of about 32 megapixels (7,680 x 4,320 image system).

Japan public broadcaster NHK - which famously initiated HD as far back as 1969 - has been also working on 7,680 x 4,320 or "Super Hi-Vision" image systems (8K.)

Super Hi-Vision standard has been recommended by ITU-R to become an international standard. It allows for programming and broadcasts at resolutions of up to 7680 by 4320, along with frame refresh rates of up to 120Hz, double that of most current HDTV broadcasts. The format also calls for a broader palette of colors that can be displayed on screen. According to specifications of SHV video for TV broadcasting, the 7,680 x 4,320 (progressive) video is provided in the 16:9 aspect ratio and is available in 120, 60 or 59.94 frames per second (FPS). Gradation is 10,12 Bit/pixel.

NHK has already succesfully tested transmissions of 8K video using terrestrial transmission technology, although for small distances.

Japan's Ministry of Internal Affairs and Communications plans to launch the test 8K TV broadcast in 2016.

Jefferson Parish close to final deal to finish Performing Arts Center ...

We're not interested in going change order by change order. We're interested in a global resolution." Jefferson Parish President John Young

Jefferson Parish is nearing a final agreement with the builder of its troubled Performing Arts Center, in a deal that could add roughly $7 million to $9 million to the tab and at least nine more months of work. That would put the center's final cost more than $25 million over budget and its timeline four years behind schedule.

Jefferson Parish President John Young and two Parish Council members confirmed Friday that the parish and LaPlace contractor Joe Caldarera are nearing a deal that would set a drop-dead date to finish the building in late 2013. Under the deal, which both sides are still negotiating, Caldarera would have to pay "substantial" fines for every day late after the deadline, officials said.

The agreement, which would require Parish Council approval, would also spell out a final payment to Caldarera to settle pending invoices for work already completed and other "justified" items still to be done. The amount of that payment, however, is still in flux.

Young declined to comment on specifics of the possible agreement. Councilmen Chris Roberts and Paul Johnston, who have been briefed on the negotiations, said Caldarera is asking for a final payment of more than $13 million dollars. But Johnston, whose district includes the Metarie location of the arts center, and Roberts both insisted the parish intends to pay less than $10 million.

"We don't want to have an almost-done facility," Roberts said. "But we're also not going to pay for unjustified charges."

"We're not going to go to $10 million," Johnston said.

Contractor wants $13 million more

Caldarera and his attorney, Joel Chaisson, were not available for comment Friday.

Roberts said the $13 million Caldarera is seeking includes a large amount in charges for delays in the project, which has been plagued by design errors and construction problems. The parish officials made it clear, however, that they don't want to pay delay costs.

When construction of the arts center began in early 2007, the project was supposed to cost $26 million and be completed by late 2009. Design errors soon complicated things, and construction problems compounded the problems. Caldarera began billing the parish for extra charges almost from the start. During early excavation to build the center's foundation, Caldarera billed the parish an extra $655,000 to clear unforeseen "underground obstructions."

The cost additions skyrocketed from there, with the administration of then-Parish President Aaron Broussard endorsing most of the extra charges and the Parish Council approving them. All along, parish officials groused about the project's rising costs and expressed hopes that no more extra charges would be added -- only to face yet another cost addition down the road. That has brought the total payments for the project to about $44 million.

Young replaced Broussard in 2010, after the former president resigned in an unrelated corruption scandal. Young said Friday that his goal is to stop the parade of cost additions, or so-called change orders, in the project. That's why, he said, his administration has not brought any new change order to the council, even as Caldarera's bills continued to pile up.

"We're not interested in going change order by change order. We're interested in a global resolution," Young said.

Parish may be short of money, again?

A key issue is how the parish will pay for that final resolution. The state last year approved $6 million in additional money for the center. The parish also recently settled its suit against designer Wisznia and Associates for $1.3 million.

But it's unclear where the parish would find the extra money if the final agreement is higher than the $7.3 million Jefferson has on hand. Roberts and Johnston said they're waiting to see any proposals from Young's administration on that issue. Young declined to discuss details, citing the ongoing negotiations.

But Young added: "Any resolution would be conditioned upon the availability of funds."

Time is pressing, also. Officials said the building doesn't have electricity and as such lacks air conditioning. That may not be a problem in winter, but could quickly damage installed sheetrock and other materials once the weather warms up in spring and summer.

"We have to get this thing behind us and get that building finished and open this year," Johnston said.

S&P lowers Illinois credit rating, blames pensions

CHICAGO (AP) ? Illinois' already disastrous financial situation worsened Friday as another credit rating agency downgraded its rating to the worst of any state in the country, blaming lawmakers' ongoing failure to resolve a multibillion-dollar pension crisis.

Standard & Poor's rating service said Friday that the rating on the state's general obligation bonds was downgraded to A- from A. The agency also gave an A- rating to $500 million in general obligation bonds that the state plans to release next week. The agency says the outlook is negative, an indication it could take the unusual step of further downgrading the state if conditions don't improve.

The downgrade is just the latest warning from the New York bond houses about the state's ongoing credit deterioration. It means taxpayers will likely pay a higher interest when the state issues bonds, or borrows money, for big items such as construction projects.

Speaking at a press conference on an unrelated topic Friday, Democratic Gov. Pat Quinn said "the pressure is higher than ever" for lawmakers to pass pension reform ? something they failed to do during a special legislative session last year and in a lame duck session that ended earlier this month, despite urgent pleas from Quinn and other leaders.

"We've got to put our seatbelts on here and understand the rating agencies won't give us better marks until the legislature passes Senate Bill 1 and gets the job done," Quinn said, referring to a recently proposed pension reform bill. "That's really the message the credit rating agencies are screaming at the top of their voice. I've heard it, and I think the members of the legislature need to hear it as well."

Illinois has a $96 billion unfunded liability in its five state-employee pension funds, due to decades of shorting or skipping its pension payments. To catch up, the state must allocate nearly one-third of its general revenue annually to pensions, putting a squeeze on money for services such as education and health care.

Standard & Poor's analysts said Friday the new rating reflects what the agency sees as the state's "weakened pension-funded ratios" and lack of action on reform measures.

"While legislative action on pension reform could occur during the current legislative session and various bills have been filed, we believe that legislative consensus on reform will be difficult to achieve given the poor track record in the past two years," analysts said.

Moody's Investors Service gave Illinois its worst rating of any state in January 2012. Earlier this month ? days after lawmakers left the lame luck session without a pension deal ? Fitch Ratings changed Illinois' financial outlook to "negative" from "stable," an indication that a ratings downgrade could be coming.

In its report Friday, Standard & Poors analysts said even if Illinois is able to pass pension legislation soon, the state is likely to face a legal challenge, so it could be years before the budget situation or the unfunded liability improve. That, along with an income tax increase that's scheduled to expire on Jan. 1, 2015, contribute to the state's negative economic outlook.

Illinois Treasurer Dan Rutherford, a Republican who's indicated he may challenge Quinn in the 2014 race for governor, said lawmakers' inaction "has our great state headed for a fiscal disaster."

"It is beyond irresponsible to let this continue," he said.

'American Horror Story' finale leaves a survivor

By Lesley Goldberg, The Hollywood Reporter

Frank Ockenfels / FX

The cast of "American Horror Story: Asylum."

When all was said and done, only one person was left standing in FX's "American Horror Story: Asylum."

The season that has featured aliens, Nazis, serial killers, demented doctors, a nun possessed by Satan, the worst insane asylum ever and?Jessica Lange?singing "The Name Game" came to a whirlwind, time-jumping conclusion when?Sarah Paulson's aspiring reporter Lana was the one to survive.

"I always thought [Lana] was the smartest cookie in the jar," co-creator/executive producer?Ryan Murphy?told reporters at a recent screening of the episode. "I liked that the hero of this season was a heroine. I like that she was a lesbian. I like that she had an arc to her sexuality. I like that she went through a lot of different things. And I like that she got a happy ending. I like that she was in a loving, accepting relationship at the end."

After jumping to the present day, the openly gay Lana is a successful reporter, best-selling author and documentarian for her work in having uncovered the misdeeds inside Briarcliff. While she's being interviewed for a Kennedy Center Honor, her son (by rape) Johnny (Dylan McDermott) is nearby awaiting an epic confrontation with his mother -- who lied that her baby with Threadson (Zachary Quinto) never survived.

Photos from THR: 'American Horror Story': The spookiest set on TV

What comes is a showdown for the ages when she reveals -- on camera -- that she lied about his death, with the duo ultimately coming face to face after the crew clears out. Revealing that he wants his late father to be proud of him, Johnny holds his mother at gunpoint. Being the smooth-talking reporter she is, Lana talks him down and reveals that Johnny isn't the monster Threadson was and then proceeds to kill the guy who posed as Bloody Face and cut off Leo's (Adam Levine) arm in the series opener.

"The thing we were most interested in writing about this season was the stuff in the last episode and the documentary series Lana made about shutting down Briarcliff. That's one of the first things when the writers landed on the idea of 'Asylum,' that period of time, those documentaries that were made," Murphy said, pointing to efforts from ?and the feature "Prophecy"?as inspiration. "It also was about the unraveling health care system in our country and how so many people were dumped there and left to rot there and all those abuses that you see. ? That was our jumping-off point. We knew we were going to have that character go in there, become a prisoner ... and go back to tear the joint down. That was the ending, which we had from the very beginning."?

UK court forces Barclays to reveal staff on Libor list

LONDON (Reuters) - A British judge forced Barclays to identify top executives alongside traders linked to a probe into rate fixing, naming ex CEOs Bob Diamond and John Varley and current Finance Director Chris Lucas on Thursday despite requests for anonymity.

The names were unveiled in a preliminary hearing for a case brought against Barclays by a residential care home operator which alleges it was mis-sold interest rate hedging products, which were based on Libor rates.

Barclays was the first bank to be punished over the Libor scandal, in which global lenders colluded to manipulate benchmark interest rates. It agreed to a fine of $453 million from U.S. and UK authorities and its then chief executive Bob Diamond left the bank following the controversy.

In the first British claim for damages, Guardian Care Homes is suing Barclays for 37 million pounds. It is seen as a test case for interest rate swaps misselling, and is also set to shine a light on people involved in the bank's manipulation of Libor and how the rate setting process was conducted.

Britain's High Court denied requests by 104 former and current Barclays staff for anonymity. The list of names includes 24 individuals who have been named in regulatory documents referring to Barclays' attempted Libor rigging. That sub list was not immediately available.

The remaining people are employees whose email accounts were disclosed to the regulatory authorities, but it is not suggested in the regulatory findings that they were implicated in the fixing scandal.

Those named include Rich Ricci, head of Barclays' investment bank.

"There is a legitimate public interest in the true picture in relation to the manipulation of Libor by banks generally, not just Barclays, being brought fully to light," the judge said in his ruling.

"In my judgment, fair and accurate media reporting of all aspects of Libor manipulation, including the involvement of employees and ex-employees of Barclays and their identity, is an important aspect of the public obtaining that true picture."

Others named on Thursday include Ryan Reich, a 30-year-old former Barclays swaps trader based in New York who was fired in 2010.

U.S. prosecutors are investigating Reich's activities while at Barclays between August 2006 and March 2010, according to several people familiar with the situation.

Ritankar "Ronti" Pal, who oversaw desk trading since 2006, was also on the list. He recently left Barclays.

Other names included Eric Bommensath, a French bond trader who became global head of fixed income and a member of the bank's executive committee, and Harry Harrison, a British banker in charge of dollar-denominated fixed-income trading in New York.

Libor is used as a benchmark for pricing trillions of dollars of loans and other financial contracts and banks implicated face a rising tide of civil lawsuits from customers who argue the rate-rigging pushed up the cost of their loans.

Switzerland's UBS agreed in December to pay fines of $1.5 billion and a slew of other banks, including Royal Bank of Scotland are also expected to reach settlements.

Barclays has said it has fired five employees following an internal investigation into how its Libor rates were submitted and disciplined another eight people. Many people identified in that investigation have also left the bank, it told lawmakers in November.

"This started as an alleged misselling case which the bank considers has no merit," Barclays said in a brief statement on Thursday. "The addition of a claim based on what happened with Libor does not change the bank's view.

"The fact that someone's documents were reviewed by the bank during its review of millions of documents does not mean that such person was involved in any wrongdoing."

Next big thing in real estate: tiny apartments

In response to skyrocketing rents in New York City, Mayor Michael Bloomberg proposed a challenge to the city's best architects: to design a space no bigger than 350 square feet into a comfortable and affordable micro-apartment. NBC's Stephanie Gosk reports.

By Ben Popken, TODAY contributor

New Yorkers are famous for their teeny apartments, but a new trend in dwelling seeks to transform those tiny spaces into big assets. They're called "micro apartments," and they make a few hundred square feet feel like over a thousand.

Fold-away beds, moveable walls, and coffee tables that expand to seat 10 for dinner are just a few of the clever touches that transform these shoe boxes into veritable mini-mansions.

"The main idea is to get double, triple, quadruple use from every space," Graham Hill, founder of the sustainable living site TreeHugger.com and the design company Life Edited, told Fair Companies?in a video interview.

With rising costs of living and a desire to limit one's environmental impact at top of mind, living simpler, and smaller, has taken off in cities around the world, especially ones known for their high rents. San Francisco recently passed an ordinance allowing for apartments to be built to 220 square feet. And "micro units" have also long been a Tokyo and Hong Kong mainstay.

For his part, Hill bought a 420-square-foot studio in Manhattan's SoHo neighborhood and?renovated?it into a concept lab for tiny living, with boutique hotel style appointments, a home theater, and a pair of drop-down bunk beds for guests.

Feeling more like 1,000+ square feet, it boasts a kitchen with top-loading fridge, freezer and dishwasher. The stove top is three induction burners stacked in a drawer. To boil a pot of water for pasta, for example, you pull the burners out and plug them in, then stow them away after the linguine is served.

Hill's design was one of 34 submitted to New York Mayor Bloomberg's?adAPT NYC contest, which tasked teams with coming up with plans for?250-370 square foot apartments designed for 1-2 person households.

The young professionals who constitute the city's lifeblood often find themselves priced out of New York City altogether or living in an outer-borough?garret.?One-third of the city's households comprise of just one individual, a number projected to rise to 46 percent in Manhattan, where one-bedrooms routinely rent for north of $2,700 a month.

New York City Mayor's Office

A rendering of the "micro unit" apartment design that won New York Mayor Bloomberg's adAPT NYC contest Tuesday.

The winning design, announced Tuesday, will form the basis for a new apartment complex built in Kip's Bay at East 27th and 1st Avenue in Manhattan with 55 of the micro units. The apartments will be built prefabricated, and then stacked on top of each other and connected - structure, plumbing, electrical, and all - like LEGOs.

New York City Mayor's Office

The design is split up into a "toolbox" and "canvas" area, with essential living amenities in the first area, and a customizable living room/bedroom slot in the second.

Called "My Micro NY," the apartment's bedroom converts into the primary living space, and the hip, 10-foot ceiling design includes a 16-foot-long overhead loft space, Juliette balconies, a?full-depth closet, full-height pull-out pantry, fridge and range.

The complex will include a garden, porch with picnic tables, lounge, laundry room, storage, bike room, and a small gym. Rents will be about $2,000 a month. Eleven of the 55 units will be reserved for households with incomes no greater than 80 percent of the area median income.

Undoubtedly there will be a waiting list to become one of the first to live in the tiny, mod apartments. If you want to see what one looks like right away though, you can visit the?Museum of New York City where a model micro-apartment is currently on display in the exhibit, "Making Room: New Models for Housing New Yorkers." You can walk around inside the unit and even try your hand at folding up and down the various pieces of hide-away furniture.

New York Mayor Bloomberg said Friday on WOR radio that he lived in an apartment for 10 years as small as the micro-units the city plans to build. His bed was a convertible couch, and he recalled it was a "pain" removing the pillows and making the bed, and not nearly as convenient as the sleek Murphy bed used in the design for the new units.

"New York?s ability to adapt with changing times is what made us the world?s greatest city ? and it?s going to be what keeps us strong in the 21st Century," said Mayor Bloomberg in a press release.

The AP contributed to this report.

Mayoral hopeful Christine Quinn proposes tablet computers for ... - Uis

BY ERIN DURKIN, NY Daily News

City schools should ditch textbooks and replace them with tablets, City Council Speaker Christine Quinn proposed Tuesday. ?Technology has changed what we need to teach, and it?s also changed how we need to teach,? the mayoral contender said in outlining a $300 million education agenda that she would implement as mayor. Quinn said the $100 million spent on textbooks every year by the school system would be enough to buy tablets for every student. She acknowledged that to stay within budget, administrators would have to find open-source texts and interactive programs for free online.

McDonald's says January restaurant sales will fall

(Reuters) - McDonald's Corp on Wednesday forecast a decline in global restaurant sales for January, as it and other fast-food chains fight for customers who are spending cautiously during continued economic uncertainty.

The world's biggest restaurant company by revenue also reported an unexpected rise in December sales at established U.S. restaurants, which helped lift its fourth-quarter profit above analysts' estimates.

Wall Street expected the early part of 2013 to be tough for McDonald's as it runs short of quick fixes for its business in the United States and bumps up against strong year-earlier results that were bolstered by unseasonably warm weather.

McDonald's forecast for a decline in this month's global same-restaurant sales suggests a "pretty clear drop off between December (2012) and January (2013)," Morningstar analyst R.J. Hottovy said.

Global same-restaurant sales were flat in December, helped by an unexpected 0.9 percent rise in the United States - its second-largest market for revenue just behind Europe.

The company's push to keep more restaurants open on Christmas Day and its shift of the limited-time offering of its popular McRib sandwich to December from October bolstered the December U.S. results. Analysts polled by Consensus Metrix had expected those sales to drop 1.78 percent.

McDonald's expects near-term top and bottom-line growth to remain pressured in part because the company must top strong results from a year ago, Chief Executive Don Thompson said on a conference call with analysts.

Last year's global sales at McDonald's restaurants open at least 13 months increased 6.7 percent for January and 7.3 percent for the first quarter.

Fast-food chains like Burger King Worldwide Inc and Yum Brands Inc's Taco Bell have introduced new U.S. menus and are doing a better job of competing with McDonald's.

At the same time, U.S. consumers have less money in their pockets since the end of the payroll tax cut.

"We suspect choppy demand trends and the impact of the loss of the payroll tax deduction (in the United States) are in part to blame" for the company's downbeat guidance, Lazard Capital Markets analyst Matthew DiFrisco said in a client note.

Fourth-quarter net income at the world's biggest restaurant chain rose 1.4 percent to $1.40 billion, or $1.38 per share. That topped analysts' average forecast of $1.33 a share, according to Thomson Reuters I/B/E/S.

Total sales rose 1.9 percent to $6.95 billion.

In a bid to remain competitive, McDonald's doubled down on promoting its value menus - such as the U.S. Dollar Menu. That effort has pressured profitability, raising concern among analysts.

Shares in the company shares were up 0.1 percent to $92.82 in early trading on the New York Stock Exchange.

Health & Fitness Tips ? Eat Healthy, Live Longer & Prevent ...

Health

It is very critical, especially as we get older, to start looking to live a healthier lifestyle. We Jamaicans and many others in the rest of the world are way too liberal when it comes to our health, lifestyle and eating habits. We don?t take care of ourselves well enough to start a balance diet and we pass on our careless ways to our children and before we know it we are dead or obese. The KFC and Burger King have to be reduced day by day and week by week. it?s very important to us to not live in the moment but to be thinking long term, whether we are 9 or 39.

Today we are bombarded with diseases and conditions; diabetes, hypertension, cancer and so many more. We need to stop the frivolous eating habits and start exercising, something most of us don?t do until a particular situation comes along. I know many people out there with diabetes. It?s a disease that?s a killer and believe it or not, prevention is better than cure and we can prevent certain types of cancer by eating right. Often times cancer chooses at random, even with the best of lifestyles, eating or otherwise.

Now we are in a world where McDonalds? or Juici Patties? or Wendy?s products are more affordable than in our parents? and grandparents? times. It?s important that we refrain from eating too much of these foods on a long term basis as it may save our lives. God knows we need to! Eating excess rice and white bread can increase the number of years we spend on this earth, take it from people who have been there and are living longer, healthier lives. They have great stories to tell.

You can?t prevent everything but you can delay them and prolong your life. Try to eat whole wheat products like bread and if you don?t like the taste try to put a bit of jam, butter or peanut butter until you can wean yourself off them and acquire the taste of the whole wheat. Don?t overdo the butter, jam or cheese as it?s not good and you can increase your fat and cholesterol intake. It should be gradual and temporary. Also, stay away from red meats and as foods you get from as before like that of Burger King or Juici Patties ore Wendy?s foods. Fried foods and greasy food are literally a recipe for death and disaster. They are serious killers so less Popeye?s and KFC!

See if you can go to the gym or job several times a week for the exercise. You?ll see great results. Eat foods like fruits or vegetables if you are fasting and don?t over eat when you break this fast. You could get bloated up. Eat less salty foods and cook with less salt as well as reduce the amount of sugary foods as you can prevent diabetes. Try a few of these tips and see how it works out for you. You would be sure to add years to your life!

German cardinal 'ashamed' at rape victim treatment

BERLIN (AP) ? A senior German bishop is publicly apologizing after two Catholic hospitals turned away a rape victim seeking treatment that included emergency contraception.

Cardinal Joachim Meisner said the hospitals should have provided the woman with medical help, though they would have drawn the line at treatment that would have prevented a pregnancy.

The incident December in the western city of Cologne caused uproar in Germany, where many hospitals are run by the Catholic and Protestant Churches.

Meisner said in a statement Tuesday that the Church was "deeply ashamed by this incident because it goes against our Christian mission."

He denied that the Church had issued an order for hospitals to turn rape victims away. He says both hospitals have also apologized.

Kars 4 Kids, American Cancer Society: Who is Most Generous To ...

Written By : Ronn Torossian
January 21, 2013

Tzedekah is a very important pillar of Judaism ? people are encouraged and required under the tenets of Judaism to give charity.??So whether its donating a car to?Kars 4 Kids?or the American Red Cross who gives the most donations:

  • Generally, Republicans give more than twice what Democrats give in charitable contributions of all sorts.??In 2008 a book was written which showed that while Democrats have higher household incomes, Republicans tend to donate more monies.
  • The Chronicle of Philanthropy?had a survey which shows that religious people of all sorts tithe due to their religious beliefs ? and then thereafter donation trends change.

o???The most generous state in the country is Utah ? where households in Provo, Utah, give away the most as a percentage ? 13.9% of discretionary income.??In second-place amongst states is Mississippi at 7.2%. Of course, Mormans ?headquarters? is in Utah ? and like Mitt Romney they tend to give generously to the Morman Church.

o???The others in the top five aren?t Morman heavy states ? but are heavy evangelical places -?Mississippi, Alabama, Tennessee and South Carolina.

One wonders indeed which party is the true kinder gentler party.??Giving is good for you.

The 7 and One Half Things You Should Know About Inauguration

A presidential inauguration is a big, long event that lasts all day and into the night-and who has time to really watch it? People have jobs, ones that don't let you off for a federal holiday.

Everyone (or, at least, some) will be talking about it, which means potential embarrassment for anyone who doesn't know what happened. Thankfully, ABC employs news professionals stationed in Washington, D.C., to pay attention to these kinds of things and boil off some of the less noteworthy or interesting stuff, presenting you with short videos of everything that really mattered. Or at least the things a lot of people were talking about.

A full day of paying attention to President Obama's second Inauguration leads one of those professionals to offer these 7 1/2 things:

1. Beyonce Sang the National Anthem

Boy, howdy! Did she ever? Beyonce has essentially become the Obama's go-to female performer: She recorded a music video for Michelle Obama's "Let's Move" initiative in 2011, and she performed at the president's last inauguration in 2009. Her velvety, soulful "Star Spangled Banner" is getting good reviews.

2. Kelly Clarkson Also Sang

Kelly Clarkson is not as "in" with the First Couple as Beyonce seems to be, but they let her sing "My Country 'Tis of Thee," and she did a pretty good job with it. This was kind of weird, though, because at one point she said she loved Ron Paul, although she later said she would vote for Obama.

3. Obama Talked About Gay Rights

This may not seem shocking since more than half the country, including President Obama, supports gay marriage. But the president made a point of mentioning gay rights during his speech, equating the struggles of the LGBT community with those of past civil rights movements, and in doing so made history.

He name-checked Stonewall, the New York City bar that was raided by police in 1969 sparking riots to protest the anti-gay crackdown. And he actually used the word "gay": "Our journey is not complete until our gay brothers and sisters are treated like anyone else under the law - for if we are truly created equal, then surely the love we commit to one another must be equal as well," Obama said in his address.

Plenty of inaugural addresses have been chock full of rhetoric about freedom and equality, but in the last four years, the political culture surrounding gay rights has changed significantly, as more states legalized same-sex marriage and as broad swaths of the country got more comfortable with homosexuality in general. Obama's "evolution" on gay marriage, and now his inaugural address, have helped signify that change.

4. Joe Biden Made Jokes and Shook Hands With People

Could we expect anything less?

Here's how the Vice President toasting Sen. Chuck Schumer instead of President Obama at the big luncheon: "I raise my glass to a man who never, never, never operates out of fear, only operates out of confidence, and a guy-I'm toasting you, Chuck." Watch it:

And here he is, scurrying around and jovially shaking hands with people along the parade route:

5. Richard Blanco Read a Poem That Was Sort of Whitman-esque, But Not Entirely

Cuban-born Richard Blanco became America's first openly gay, Latino Inauguration poet. He read a nine-stanza poem entitled "One Today," which set a kind of unifying American tableau scene.

6. Obama and Michelle Walked Around Outside The Limo

President Obama walked part of the parade route, from the Capitol to the White House, with Michelle. They waved to people. It is not entirely abnormal for a president to do this at an inaugural parade. But they walked quite a ways.

7. John Boehner: 'Godspeed'

The speaker of the House presented American flags to Obama and Biden, telling them: "To you gentlemen, I say congratulations and Godspeed."

7 1/2. Sasha and Malia Were There.

Obama's daughters, Sasha and Malia, were there. They didn't really do much, but they did wear coats of different shades of purple that got a lot of attention on Twitter.

Reports of the daughters looking at smartphones and applying lip gloss highlighted their day. As did this .gif of Sasha yawning.
